Overview

Ad-BSP-E1a is a conditionally replicating oncolytic adenovirus engineered with the bone sialoprotein (BSP) promoter to drive the expression of the adenoviral E1A gene[3][4][1]. This design confers tumor specificity, restricting viral replication to BSP-expressing cells, which are highly abundant in the extracellular matrix of bone and are secreted by osteoblasts and osteoclasts. BSP is overexpressed in several cancer types, especially those with bone involvement, such as prostate cancer, breast cancer, lung cancer, and melanoma[3]. In prostate cancer models—including androgen-independent intraosseous settings—Ad-BSP-E1a exhibits selective replication and oncolytic activity in tumor cells, with limited activity in normal tissues[2][4]. The E1A gene product is crucial for early viral gene activation, disruption of cell cycle regulation via retinoblastoma (Rb) pathway inhibition, immune evasion, and apoptosis[5]. Preclinical data show Ad-BSP-E1a effectively inhibits tumor growth and reduces PSA levels in vitro and suppresses subcutaneous and intraosseous xenograft tumor growth in vivo[3].
