**Ad-CAIX** is a replication-defective adenoviral (Ad) vaccine that encodes the tumor antigen **carbonic anhydrase IX (CAIX)**. The drug is designed as an immunotherapy approach for cancer, especially renal cell carcinoma. Ad-CAIX works by using the adenovirus as a vector to deliver the CAIX antigen to the host, thereby stimulating a potent **anti-tumor immune response** primarily through the activation of CD8+ cytotoxic T cells, increased maturation of dendritic cells, and enhanced infiltration of these immune cells into the tumor microenvironment. The vaccine aims to induce tumor-specific immune responses and has demonstrated significant suppression of tumor growth, reduction in tumor volume, and prevention of metastasis in preclinical renal carcinoma models. The strategy leverages the ability of the adenovirus to act as an adjuvant, enhancing the immunogenicity of the otherwise weak CAIX antigen. Ad-CAIX can be used as a single agent or in combination with other vector-based immunotherapies targeting immune checkpoints[1][8][2].
