Ad-CAIX + Ad-PD-L1 is an investigational combination vaccine therapy developed by researchers at Xuzhou Medical University for the treatment of renal cell carcinoma. This gene therapy-based approach consists of two separate replication-defective adenovirus (Ad5) vectors administered intramuscularly. Ad-CAIX encodes the tumor-associated antigen Carbonic Anhydrase IX (CAIX), which is highly expressed on the surface of clear cell renal cell carcinoma (ccRCC) cells. Ad-PD-L1 encodes the immune checkpoint protein Programmed Death-Ligand 1 (PD-L1). The dual-targeting strategy aims to generate a robust anti-tumor immune response by directing the immune system to attack CAIX-expressing tumor cells while simultaneously targeting PD-L1 to overcome tumor-induced immunosuppression. Preclinical studies have demonstrated that this combination vaccine enhances dendritic cell maturation and promotes multifunctional CD8+ T-cell-mediated immune responses, effectively inhibiting primary tumor growth and lung metastasis in animal models.
