Overview

Ad-CD40L is a **gene therapy** product consisting of a non-replicating or tumor-restricted replication-competent **adenoviral vector** encoding the human **CD40 ligand (CD40L, also known as CD154)**. The therapy is designed to induce local expression of CD40L within tumors, primarily **diffuse midline glioma** (formerly known as diffuse intrinsic pontine glioma, DIPG) and other brainstem gliomas. CD40L is an immune co-stimulatory molecule that binds to CD40 on antigen-presenting cells, leading to immune activation, infiltration of CD4+ T cells and other effector immune cells, and induction of anti-tumor immunity. Experimental and translational data show that Ad-CD40L can recruit and activate both innate and adaptive immune responses, with CD4+ T cells being especially critical for efficacy. Tumor-restricted, replication-competent versions (rAd-Δ24-CD40L) are being explored preclinically and are designed to limit toxicity by restricting viral replication and CD40L expression to tumor cells[1][5][8].
