Ad-COX2-E1a is a **replication-selective adenoviral vector** in which the expression of the adenoviral E1a gene—required for viral replication—is controlled by the **cyclo-oxygenase 2 (COX-2) promoter**. The drug is designed for **oncolytic therapy**: it selectively replicates in and destroys tumor cells that express COX-2, such as squamous cell carcinoma of the head and neck[1]. By driving E1a expression with the COX-2 promoter, replication occurs preferentially in COX-2–positive cells, limiting activity in normal tissues.
