**AD-DELO3-RGD** is a genetically engineered, YB-1-dependent oncolytic adenovirus developed to selectively replicate in and lyse cancer stem cells (CSCs), especially those found in glioblastoma (GBM). The virus is modified to replicate efficiently only in cells exhibiting high expression of the YB-1 transcription factor, which is linked to tumor progression, therapy resistance, and poor prognosis. The RGD modification refers to the insertion of a peptide motif (Arg-Gly-Asp) in the adenoviral fiber knob, enhancing infection of tumor cells via integrin receptors highly expressed on CSCs and tumor cells. Mechanism of action: AD-DELO3-RGD enters tumor cells via integrin receptors, selectively replicates in YB-1-positive cells, and induces direct oncolysis. Replication and cytolysis are markedly inhibited in normal brain cells with low YB-1 expression, increasing the safety index. Preclinical studies show potent tumor inhibition in TMZ-resistant GBM models and significant prolongation of survival in animal models[1]. Developed primarily for the treatment of glioblastoma and potentially other tumors characterized by cancer stem cells with high YB-1 expression.
