The Ad-ΔLMP1-LMP2 DC vaccine is an experimental autologous dendritic cell (DC) immunotherapy designed to treat Epstein-Barr virus (EBV)-positive nasopharyngeal carcinoma (NPC). The vaccine consists of a patient's own dendritic cells that have been genetically modified using a recombinant adenovirus vector to express two specific EBV antigens: a truncated version of Latent Membrane Protein 1 (ΔLMP1) and the full-length Latent Membrane Protein 2 (LMP2). By presenting these viral antigens, the vaccine aims to stimulate a T cell-mediated immune response against NPC cells that express these proteins. Clinical evaluation in Phase II studies has demonstrated safety and the induction of delayed-type hypersensitivity responses, although clinical efficacy in advanced metastatic disease remains a focus for further optimization.
