Overview

Ad-G-AT2R-eGFP is a recombinant adenoviral gene therapy vector designed to deliver and overexpress the human angiotensin II type 2 receptor (AT2R). The vector construct contains the genomic DNA of AT2R, including introns 1 and 2 and the encoding region, along with an enhanced green fluorescent protein (eGFP) reporter gene, both driven by cytomegalovirus (CMV) promoters. In preclinical studies, particularly in bladder cancer (BCa), prostate cancer, and hepatocellular carcinoma, the overexpression of AT2R mediated by this vector has demonstrated significant anti-tumor activity. The mechanism of action involves the induction of apoptosis through the activation of p38 MAPK and the extrinsic signaling pathway (caspase-8 and caspase-3), while simultaneously inhibiting cell proliferation and angiogenesis by downregulating ERK MAPK and vascular endothelial growth factor (VEGF) expression.
