Overview

Ad-hOC-E1 is an oncolytic adenovirus engineered for the treatment of advanced renal cell carcinoma (RCC). Its replication is transcriptionally regulated by the human osteocalcin (hOC) promoter, which is transcriptionally silent in most tissues but highly inducible by Vitamin D3. This design allows for a 'pharmaceutically-controlled' replication strategy. In preclinical studies, the virus is loaded into human bone marrow-derived mesenchymal stem cells (hMSCs), which act as carriers that home to tumor sites in response to PDGF-AA chemoattraction. Upon reaching the tumor, administration of Vitamin D3 induces the hOC promoter to drive the expression of the viral E1 gene, triggering viral replication and the subsequent lysis of RCC cells. This approach aims to improve the safety and efficacy of systemic oncolytic virotherapy by restricting viral activity to the tumor microenvironment and utilizing cell-based delivery to bypass immune clearance.
