Ad-IL-1β is a replication-deficient recombinant adenoviral vector (typically based on human adenovirus serotype 5, Ad5) that encodes the mature form of interleukin-1 beta (IL-1β). Developed primarily as a genetic adjuvant for mucosal vaccines, Ad-IL-1β is co-administered intranasally or via other mucosal routes alongside antigen-expressing vectors (such as those targeting respiratory syncytial virus or influenza) to enhance local immune responses. It functions by expressing IL-1β, which recruits innate and adaptive immune cells, promotes dendritic cell migration, and stimulates the generation of antigen-specific secretory immunoglobulin A (IgA) and tissue-resident memory T cells (TRM) in the respiratory tract. Additionally, Ad-IL-1β has been utilized in preclinical research models of airway remodeling, chronic obstructive pulmonary disease (COPD), and pulmonary metastasis to study IL-1β-mediated inflammation, tissue fibrosis, and local antitumor immunity.
