Ad-MAGEA3 is a recombinant adenovirus type 5-based cancer vaccine engineered to express the human melanoma antigen A3 (MAGE-A3), a tumor-associated antigen found in various cancers but not in normal tissues. Upon administration, the vaccine selectively infects and replicates within cancer cells, leading to expression of MAGE-A3 and induction of an anti-tumor immune response, particularly by priming CD8+ T-cells against MAGE-A3 epitopes. It is primarily being investigated as part of a prime-boost immunization strategy with MG1-MAGEA3 (an oncolytic Maraba virus expressing MAGE-A3) for the treatment of solid tumors such as non-small cell lung cancer (NSCLC). The approach aims to stimulate robust cellular immunity against tumors expressing the MAGE-A3 antigen[1][2][3][8].
