**Ad-MG1-MAGEA3** is a prime-boost cancer vaccine regimen combining Ad-MAGEA3, an adenovirus vector expressing the MAGE-A3 tumor antigen, with MG1-MAGEA3, an oncolytic Maraba MG1 virus also expressing MAGE-A3. The Ad-MAGEA3 primes CD4+ and CD8+ T-cell responses intramuscularly, while the subsequent intravenous MG1-MAGEA3 boost expands these antigen-specific T-cells (up to >2% of circulating CD8+ T-cells), induces humoral immunity with anti-MAGE-A3 antibodies, and leverages oncolytic activity to reprogram the tumor microenvironment for enhanced innate and adaptive antitumor immunity. Developed originally by researchers at McMaster University and advanced by Turnstone Biologics (formerly part of Agenus), it targets MAGE-A3-positive solid tumors such as melanoma, NSCLC, and others, with preclinical safety in nonhuman primates and phase 1/2 trials demonstrating tolerability and immunogenicity when combined with pembrolizumab (NCT02285816, NCT02879760, NCT03773744).[1][3][7][8][9][15]
