Overview

Ad-MMP-2-Si is an adenoviral vector-based gene therapy designed to deliver small interfering RNA (siRNA) targeting matrix metalloproteinase-2 (MMP-2). MMP-2, also known as gelatinase A, is a zinc-dependent endopeptidase involved in the degradation of the extracellular matrix, which is a critical step in tumor invasion and metastasis. In glioblastoma models, Ad-MMP-2-Si has been shown to downregulate MMP-2 expression, leading to the inhibition of cell migration and invasion. This effect is mediated through the disruption of the MMP-2/integrin αVβ3/FAK signaling complex, which subsequently reduces the activation of Rho GTPases Rac1 and Cdc42. The therapy was primarily investigated in preclinical settings for the treatment of malignant gliomas.
