Overview

AD-O21.1 is a recombinant fusion protein variant of TRAIL (TNF-related apoptosis-inducing ligand), also known as Apo2L, developed by Adamed Pharma (including its subsidiary Polfa Pabianice). The drug is designed to selectively induce apoptosis in cancer cells by acting as an agonist for the death receptors DR4 (TNFRSF10A) and DR5 (TNFRSF10B). Unlike first-generation recombinant TRAIL molecules, AD-O21.1 is a modified fusion variant engineered to enhance stability, improve pharmacokinetic properties, and increase pro-apoptotic activity within the tumor microenvironment. It is currently being evaluated in Phase 1 clinical trials for the treatment of advanced solid tumors, where it aims to overcome the therapeutic limitations observed with native TRAIL proteins.
