Overview

AD-O51.4 is a recombinant fusion protein developed by Adamed Pharma S.A. for the treatment of advanced solid tumors. It is a modified version of the TNF-related apoptosis-inducing ligand (TRAIL), engineered to improve upon the pharmacokinetic and stability issues encountered by earlier TRAIL-based therapies. The drug acts as an agonist for death receptors 4 (DR4) and 5 (DR5), which are frequently overexpressed on the surface of cancer cells. Upon binding, AD-O51.4 activates the extrinsic apoptotic pathway, leading to selective programmed cell death in malignant tissues while sparing healthy cells. It is currently being evaluated in Phase 1 clinical trials to determine its safety, tolerability, and preliminary efficacy in patients with refractory cancers.
