Ad-p16 is an investigational gene therapy in which an adenoviral vector (Ad) delivers the human p16 gene to target cells, aiming to restore or enhance expression of p16, a cyclin-dependent kinase inhibitor with tumor suppressor activity. The p16 protein slows cell division by inhibiting cell cycle progression from G1 to S phase, making it a critical regulator of cellular senescence and a tumor suppressor implicated in a range of cancers. Ad-p16 is primarily being studied for cancers where CDKN2A (the gene encoding p16) is inactivated or its product is deficient, with the intended mechanism to restore the tumor-suppressive cell cycle arrest in malignant cells[1][3][5].
