Overview

Ad-p27 is a **gene therapy vector** comprising a recombinant adenovirus engineered to express the cyclin-dependent kinase inhibitor p27Kip1 (CDKN1B), a tumor suppressor protein that negatively regulates cell cycle progression by inhibiting cyclin-CDK complexes. The overexpression of p27Kip1 via Ad-p27 induces cell cycle arrest, promotes apoptosis, and reduces proliferation in cancer cells. In preclinical studies, intratumoral administration of Ad-p27 led to tumor regression and increased apoptosis in xenograft tumor models. The vector is primarily investigated as a potential therapeutic for cancer, notably breast and lung cancer, based on its ability to restore or bolster p27Kip1 activity where it is deficient.
