Overview

Ad-p53 is a gene therapy product developed by MultiVir, Inc. for the treatment of various solid tumors, including colorectal cancer, hepatocellular carcinoma, and head and neck squamous cell carcinoma. It utilizes a replication-impaired adenoviral vector to deliver a functional copy of the human p53 tumor suppressor gene (TP53) directly into tumor cells. The p53 protein is a master regulator of the cell cycle and apoptosis; its restoration in p53-deficient or mutated tumors is intended to inhibit cell proliferation, induce programmed cell death, and sensitize the tumor microenvironment to other therapies such as chemotherapy and immune checkpoint inhibitors. Ad-p53 is typically administered via intra-arterial or intra-tumoral injection and is currently being investigated in Phase 1/2 clinical trials in combination with agents like capecitabine, pembrolizumab, and nivolumab to overcome treatment resistance in advanced or metastatic cancers.
