Ad-p95-300 is a recombinant, replication-deficient adenoviral vector designed for the treatment of head and neck squamous cell carcinoma (HNSCC). It functions as a radiosensitizer by delivering a truncated version of the *NBN* gene, which encodes the C-terminal 300 amino acids of the Nbs1 (p95) protein. This mutant protein acts in a dominant-negative fashion to disrupt the formation and function of the MRN complex (Mre11-Rad50-Nbs1), a critical component of the cellular DNA double-strand break repair machinery. By inhibiting DNA repair, Ad-p95-300 enhances the cytotoxic effects of ionizing radiation, induces G2/M cell cycle arrest, and reduces telomerase activity in tumor cells. The therapy has demonstrated efficacy in preclinical in vitro models of HNSCC, showing significant antitumor effects both as a monotherapy and in combination with radiation.
