Ad-PLCη2 is an adenoviral gene therapy vector designed to overexpress Phospholipase C eta 2 (PLCη2) or its specific C2 domain (PLCη2_C2). It is currently being investigated for the treatment of rheumatoid arthritis (RA). The therapeutic mechanism involves the PLCη2 C2 domain binding to HRAS, which disrupts the HRAS-RAF1 protein-protein interaction. This disruption leads to the suppression of downstream proinflammatory signaling pathways, including the NF-κB, JAK-STAT, and MAPK pathways. In preclinical models, Ad-PLCη2 has demonstrated the ability to inhibit the tumor-like phenotype of fibroblast-like synoviocytes (FLSs), reducing their proliferation, migration, invasion, and inflammatory cytokine production, thereby mitigating joint destruction and arthritis pathology.
