AD-PluReceptor-NK cells are an investigational allogeneic natural killer (NK) cell therapy developed at the MD Anderson Cancer Center. These cells are engineered using the proprietary "PluReceptor" (Plug-and-Play) platform, which features a universal high-affinity chimeric receptor, typically an engineered version of the Fc gamma receptor IIIa (CD16a/FCGR3A). This modular design allows the NK cells to be "plugged" into various monoclonal antibodies, effectively redirecting their cytotoxic activity toward specific antigens via antibody-dependent cellular cytotoxicity (ADCC). In current clinical studies for autoimmune disorders, these cells are being evaluated in combination with the anti-CD19 monoclonal antibody tafasitamab to achieve deep depletion of B cells. The cells are often further modified to express cytokines like IL-15 to enhance their survival, expansion, and persistence in the recipient's circulatory system.
