The Ad-PR vaccine is a preclinical, first-generation adenoviral vector-based cancer vaccine encoding the progesterone receptor (PR), specifically isoforms PR-A and PR-B. Developed by researchers at Duke University and the University of Kansas, the vaccine is designed to immunologically target hormone receptor-positive (HR+) breast cancer. Progesterone receptor signaling has been shown to play an immunosuppressive role in the tumor microenvironment of HR+ breast cancers, contributing to resistance against endocrine inhibition therapy. By delivering codon-optimized human or mouse PR sequences via an adenoviral vector, the vaccine elicits PR-specific T-cell responses (specifically CD8+ cytotoxic T cells) to overcome local immunosuppression and target PR-expressing tumor cells.
