Overview

Ad-Relaxin (also known as GM101) is an engineered oncolytic adenovirus designed to express the human relaxin transgene, primarily developed for the treatment of advanced solid tumors with high levels of fibrosis. The therapeutic strategy utilizes the oncolytic properties of the adenovirus to selectively infect and lyse cancer cells, while the expressed relaxin protein works to remodel the tumor microenvironment. Relaxin facilitates the degradation of the tumor extracellular matrix (ECM) by inducing the expression of matrix metalloproteinases (MMPs), which break down collagen and other structural components that typically hinder drug delivery. By reducing intratumoral pressure and physical barriers, Ad-Relaxin enhances the dispersion of the virus itself and improves the penetration of co-administered therapies, such as chemotherapy or immune checkpoint inhibitors like anti-PD-1 antibodies.
