Ad-RTS-mIFNα is a preclinical recombinant adenoviral gene therapy vector designed for the conditional, regulated expression of murine interferon-alpha (mIFNα). It utilizes the RheoSwitch Therapeutic System (RTS), an inducible promoter system that allows precise control over transgene expression through the administration of an oral small-molecule activator ligand, such as veledimex (INXN-1001). Developed by Intrexon (now Precigen), Ad-RTS-mIFNα has been evaluated in preclinical syngeneic mouse models of cancer, including Lewis lung carcinoma (LLC) and mammary carcinoma (4T1), both as a monotherapy and in combination with Ad-RTS-mIL-12, demonstrating significant tumor growth inhibition and anti-tumor immune activation without systemic toxicity.
