Ad-SGE-REIC-NS is an investigational gene therapy developed by Momotaro-Gene Inc. for the treatment of recurrent malignant glioma. It utilizes a non-replicating adenovirus vector to deliver the Reduced Expression in Immortalized Cells (REIC)/Dickkopf-3 (DKK3) gene, a tumor suppressor that is frequently downregulated in various malignancies. The SGE (Super Gene Expression) component refers to a specialized promoter system designed to significantly enhance the expression levels of the therapeutic gene within target cells. Upon delivery, the overexpression of REIC/DKK3 protein triggers endoplasmic reticulum (ER) stress-mediated apoptosis specifically in cancer cells while sparing normal cells. Furthermore, the therapy induces a bystander effect and stimulates systemic anti-tumor immunity by promoting the maturation of dendritic cells and the activation of cytotoxic T lymphocytes. Ad-SGE-REIC-NS is currently being evaluated in Phase I/II clinical trials, representing a second-generation advancement over the earlier MTG-201 (Ad-REIC) candidate.
