Overview

Ad-shTGF-beta2 is an experimental gene therapy consisting of a recombinant, replication-deficient adenovirus vector (typically Adenovirus type 5) engineered to deliver a short hairpin RNA (shRNA) expression cassette targeting the messenger RNA (mRNA) of Transforming Growth Factor Beta 2 (TGF-beta 2). TGF-beta 2 is a potent immunosuppressive cytokine that is frequently overexpressed in the tumor microenvironment, where it facilitates immune evasion, promotes angiogenesis, and drives epithelial-mesenchymal transition (EMT). Upon transduction of target cells, the shRNA is processed by the endogenous RNA interference (RNAi) machinery into small interfering RNAs (siRNAs) that mediate the sequence-specific degradation of TGF-beta 2 mRNA, thereby reducing protein levels. This knockdown is intended to restore anti-tumor immune responses, particularly by enhancing T-cell and natural killer (NK) cell activity. Beyond oncology, Ad-shTGF-beta2 has been investigated for fibrotic conditions such as hypertrophic scarring, where it aims to inhibit excessive fibroblast activation and collagen deposition. It has been evaluated in early-phase clinical trials for advanced solid tumors and hypertrophic scars.
