Ad-siSSF is a gene therapy-based immunotherapeutic agent utilizing an adenoviral vector to deliver a combination of immunomodulatory payloads. Specifically, Ad-siSSF encodes for two tumor-associated antigens, survivin and MUC1, to enhance tumor-specific immune recognition, secretory bacterial flagellin to promote dendritic cell (DC) maturation, and an RNA silencing component targeting intracellular immune checkpoints. This strategy is designed to elicit a robust anti-tumor immune response by both directly activating dendritic cells and reducing tumor-induced immune suppression within the tumor microenvironment. The agent is evaluated as part of a genetically modified DC vaccine platform for cancer immunotherapy[5][7].
