Ad-siSSF-DC is an adenovirus-based genetically modified dendritic cell (DC) vaccine developed by the Affiliated Hospital to Academy of Military Medical Sciences for the treatment of acute myeloid leukemia (AML). The vaccine utilizes an adenoviral vector (Ad-siSSF) to engineer monocyte-derived dendritic cells to express three functional components: a survivin-MUC1 fusion protein serving as tumor-associated antigens (TAAs), a SOCS1 short hairpin RNA (shRNA) that acts as an intracellular immune checkpoint inhibitor to override the SOCS1 molecule, and secretory bacterial flagellin which serves as a Toll-like receptor 5 (TLR-5) agonist to induce DC maturation. This biologic immunotherapy is designed to stimulate a potent anti-leukemic T-cell response and is frequently evaluated in clinical trials in combination with cytokine-induced killer (CIK) cells.
