Overview

Ad-TD-nsIL12 is an oncolytic adenovirus engineered to deliver a modified version of interleukin-12 (IL-12). It's designed to selectively replicate in tumor cells and express a non-secretory form of IL-12 that remains confined to the tumor microenvironment, limiting systemic toxicity. The virus is based on adenovirus serotype 5 (Ad5) but has three gene deletions (E1A-CR2, E1B19K, and E3gp19K) and uses the E3gp19K promoter to control IL-12 expression. When infected tumor cells lyse, they release both IL-12 and tumor-associated antigens, creating a synergistic effect that induces an efficient anti-tumor immune response. This response can target distant tumor cells, including metastatic lesions not directly infected by the virus.
