Ad-TEM8 is a preclinical-stage **DNA-based vaccine** targeting **tumor endothelial marker 8 (TEM8/ANTXR1)**, which is implicated in tumor angiogenesis and progression. Ad-TEM8 works by encoding the extracellular domain of the TEM8 protein, aiming to stimulate an immune response against TEM8-positive tumor vasculature and tumor cells. The vaccine has demonstrated antitumor effects in preclinical murine models, enhancing inflammation and tumor necrosis through CD8+ T cell activation, particularly when used in combination with other DNA vaccines targeting different antigens[1]. TEM8 is highly expressed in several human cancers (including breast, colorectal, lung, prostate, pancreatic, gastric, and ovarian cancer), and its high expression is associated with more aggressive disease and poor prognosis[1][4]. Ad-TEM8 is one among several emerging strategies to therapeutically target TEM8 in the tumor microenvironment.
