**Ad-tk-MCP1** is an experimental bicistronic recombinant adenoviral gene-therapy vector engineered to express herpes simplex virus thymidine kinase and human monocyte chemoattractant protein-1 from a CAG-promoter-driven cassette separated by an encephalomyocarditis-virus internal ribosomal entry site. In hepatocellular carcinoma models, tumor-directed administration followed by ganciclovir activates the herpes simplex virus thymidine kinase suicide-gene system, while monocyte chemoattractant protein-1 promotes macrophage recruitment and macrophage-associated antitumor activity, including enhanced tumor necrosis factor-alpha production. The construct showed enhanced antitumor activity in subcutaneous hepatocellular carcinoma xenograft models compared with vectors expressing either transgene alone or with the reverse transgene order.
