**Ad-Tp-E1A-Gp-p53** is an experimental, conditionally replicative oncolytic adenovirus engineered for glioma-directed therapy. The construct uses the human telomerase reverse transcriptase promoter to drive adenoviral E1A expression and preferential viral replication in tumor cells, while an enhanced glial fibrillary acidic protein promoter drives expression of a therapeutic wild-type p53 transgene. In a 2017 in-vitro study, the virus increased p53 expression and inhibited growth of U251 glioma cells in a multiplicity-of-infection-dependent manner. It is a preclinical research construct rather than an approved medicine.
