Ad-VT is a **dual-specific oncolytic adenovirus** engineered for tumor-specific targeting and killing. The drug contains the E1a gene driven by the human telomerase reverse transcriptase promoter (hTERTp), active in most human cancers but not in normal cells, and expresses **apoptin (VP3 protein)** from chicken anemia virus, which selectively induces apoptosis in tumor cells. Ad-VT has demonstrated the ability to inhibit proliferation, induce apoptosis and autophagy, block cell cycle progression, suppress angiogenesis, and disrupt cell energy metabolism in chemotherapy-resistant lung cancer cells. It also downregulates P-glycoprotein (P-gp) and inhibits the PI3K/Akt/mTOR pathway, enhancing chemosensitivity, especially in gemcitabine- and paclitaxel-resistant non-small cell lung cancer[1].
