Ad-ZBP-89 is a replication-deficient recombinant adenoviral vector designed to express the full-length human ZBP-89 (Zinc-binding protein-89) cDNA. ZBP-89 is a Krüppel-type zinc-finger transcription factor that plays a critical role in regulating cell growth and apoptosis. In preclinical studies, particularly for hepatocellular carcinoma (HCC), Ad-ZBP-89-mediated overexpression of ZBP-89 has been shown to induce apoptosis through both p53-dependent and p53-independent pathways. The mechanism involves the upregulation of pro-apoptotic proteins such as Bak and the downregulation of anti-apoptotic factors, often through epigenetic modulation including the inhibition of histone deacetylase 3 (HDAC3) and DNA methyltransferase 1 (DNMT1). While it serves as a potent research tool for investigating gene therapy approaches in gastrointestinal cancers, it is not currently a marketed pharmaceutical product.
