Overview

Ad.asHsp70 is a recombinant adenoviral vector expressing antisense heat shock protein 70 (Hsp70) cDNA. Developed by researchers at the Danish Cancer Society, it is designed to selectively deplete Hsp70 in tumor cells. Hsp70 is an antiapoptotic chaperone protein that is highly expressed in human tumors and protects them from lysosomal membrane permeabilization and cell death. Depletion of Hsp70 by Ad.asHsp70 triggers lysosomal membrane permeabilization, leading to the release of lysosomal cysteine proteases (cathepsins) into the cytosol and inducing massive, caspase-independent, tumor-specific cell death. In preclinical models, locoregional administration of Ad.asHsp70 has demonstrated efficacy in eradicating xenografts of glioblastoma, breast carcinoma, and colon carcinoma.
