The Ad.p53-DC vaccine is a dendritic cell-based therapeutic vaccine designed to activate the immune system against cancer cells that express high levels of p53. This investigational therapy combines the patient's own dendritic cells with a genetically engineered adenovirus carrying the human p53 gene. The vaccine is created by collecting dendritic cells from the cancer patient and infecting them in the laboratory with an adenovirus vector carrying the human wild-type p53 gene (Ad-p53). These modified dendritic cells are then injected back into the patient, typically via subcutaneous administration[3][6]. The underlying principle is that these modified dendritic cells will present p53 antigens to the immune system, stimulating a specific immune response against tumor cells that express mutant p53 proteins. This is particularly relevant since p53 gene mutations and protein overexpression are present in approximately 90% of small cell lung cancer (SCLC) cases and many other cancer types[2].
