Overview

Ad.Rz.GFP is a replication-deficient bicistronic adenoviral vector designed for gene therapy in cancer. It coexpresses green fluorescent protein (GFP) and a hammerhead ribozyme (such as Rz184 or Rz305) that specifically targets and degrades human stathmin (STMN1) mRNA. Stathmin is a microtubule-destabilizing phosphoprotein overexpressed in various malignancies, including breast and prostate cancers, where it plays a critical role in cell cycle progression and mitotic spindle regulation. By downregulating stathmin expression, Ad.Rz.GFP inhibits tumor cell proliferation, disrupts the mitotic apparatus, and induces apoptosis. It has been investigated in preclinical models of breast cancer and prostate cancer, both as a monotherapy and in synergistic combination with chemotherapeutic agents like paclitaxel.
