Ad.TRAIL (also referred to as Ad5-TRAIL or Ad-TRAIL) is a replication-deficient recombinant adenoviral gene therapy vector (specifically adenovirus serotype 5) encoding the human tumor necrosis factor-related apoptosis-inducing ligand (TRAIL) cDNA. Developed by researchers at the University of Iowa, Ad.TRAIL is designed to deliver the TRAIL gene directly into tumor cells, leading to the expression of membrane-bound TRAIL protein on the cell surface. This protein selectively binds to death receptors 4 and 5 (DR4 and DR5) on cancer cells, triggering the extrinsic apoptotic pathway and inducing tumor cell death while sparing normal tissues. Ad.TRAIL has been evaluated in a Phase I clinical trial for the treatment of locally-confined prostate cancer and has shown potential in preclinical studies for other malignancies, including renal cell carcinoma, pancreatic cancer, and non-small cell lung cancer.
