Overview

AD16 is a novel small molecule drug candidate under development for the treatment of Alzheimer's disease. It acts as an inhibitor of LINE-1 retrotransposon endonuclease, with an IC50 of 4.7 μM, and has demonstrated potential benefits in preclinical studies by reducing neuroinflammation—a key pathological feature in Alzheimer's disease[1][2]. In early clinical trials involving healthy adults, AD16 showed favorable safety, tolerability, and pharmacokinetic profiles. The drug is orally administered and its absorption rate can be affected by food intake (specifically high-fat meals), though overall absorption remains unchanged[1]. No serious or dose-related toxicities were observed in these studies.
