Ad26.CS.01 is an experimental viral vector vaccine developed for the prevention of malaria. It utilizes a replication-deficient recombinant adenovirus serotype 26 (Ad26) vector to deliver the Plasmodium falciparum circumsporozoite (CS) protein, which is the primary surface protein of the malaria parasite during the sporozoite stage. The vaccine is produced using the proprietary AdVac technology and PER.C6 cell line. It is designed to stimulate both humoral and cellular immune responses to prevent the parasite from infecting liver cells. In clinical development, Ad26.CS.01 has been evaluated as a booster component in heterologous prime-boost regimens, specifically following priming with Ad35.CS.01. Although Phase 1/2a trials confirmed the vaccine's safety and immunogenicity, it demonstrated limited protective efficacy against controlled human malaria infection.
