Drug intelligence / Profile preview
Ad26.Mos4.HIV + clade C gp140 is a **combination HIV vaccine regimen** consisting of a recombinant adenovirus serotype 26 (Ad26) vector vaccine that expresses four mosaic HIV-1 antigens (Ad26.Mos4.HIV) and a recombinant trimeric HIV-1 envelope glycoprotein protein from clade C (gp140), adjuvanted with aluminum phosphate. The Ad26.Mos4.HIV component contains four mosaic immunogens encoded by Ad26 vectors: Mos1.Gag-Pol, Mos2.Gag-Pol, Mos1.Env, and Mos2S.Env, designed for broader immunological coverage of globally circulating HIV-1 strains. The clade C gp140 is a recombinant trimeric glycoprotein derived from the HIV-1 Env protein (strain 97ZA012). This combination is typically administered as a prime-boost regimen with the goal to induce broad humoral and cellular immune responses against multiple HIV-1 clades. The vaccine regimen has been developed and tested primarily for the **prevention of HIV-1 infection** in at-risk populations, including men who have sex with men, transgender individuals, and women in settings with high HIV prevalence. The regimen has been evaluated in large-scale efficacy trials such as Imbokodo (HVTN 705) and Mosaico for its ability to generate binding and neutralizing antibodies and T cell responses, but did not demonstrate statistically significant efficacy and has been discontinued for advanced clinical development[1][2][3][4][5][6][7].
