Ad26.VP22.7Epi is an experimental cancer vaccine candidate that utilizes an adenovirus serotype 26 (Ad26) vector to deliver tumor-specific neoantigens. The construct is specifically engineered to encode seven neoantigen epitopes (7Epi) and incorporates the herpes simplex virus type 1 (HSV-1) VP22 protein, which is used to enhance antigen presentation and promote intercellular transport. Developed by researchers at Beth Israel Deaconess Medical Center and Harvard Medical School, the vaccine is designed to induce robust, hyperexpanded CD8+ T cell responses within the tumor microenvironment. In preclinical mouse models (B16-F10 and MC38), Ad26.VP22.7Epi demonstrated superior protective efficacy and T cell maturation compared to traditional adjuvanted peptide vaccines, specifically targeting neoantigens such as Adpgk and Reps1.
