Overview

Ad2CMV-MART1 is an investigational therapeutic cancer vaccine and gene therapy developed by the National Cancer Institute's Surgery Branch in collaboration with Genzyme. It utilizes a replication-defective adenovirus vector to deliver the gene encoding the melanoma antigen recognized by T-cells 1 (MART-1), also known as Melan-A. Upon administration, the viral vector enters host cells and induces the expression of the MART-1 protein, which is then processed and presented on the cell surface. This presentation is intended to stimulate a specific cytotoxic T-lymphocyte (CTL) response against melanoma cells that naturally express the MART-1 antigen. While Phase I clinical trials demonstrated the vaccine's safety at high doses, its efficacy was potentially limited by pre-existing neutralizing antibodies against the adenovirus vector in many patients.
