Ad35-ENV is a recombinant, replication-defective adenovirus serotype 35 (Ad35) viral vector vaccine developed for the prevention of HIV infection. Created through a collaboration between the International AIDS Vaccine Initiative (IAVI) and Janssen (a subsidiary of Johnson & Johnson), the vaccine utilizes Janssen's proprietary AdVac technology. The vector is engineered to carry and express the envelope (env) genes derived from HIV-1 subtype A isolates. Its primary mechanism of action is to induce HIV-specific cellular and humoral immune responses against the HIV envelope protein. In clinical development, Ad35-ENV has been evaluated in Phase 1 trials, often administered as part of a heterologous prime-boost regimen in combination with other viral vectors, such as Ad26.ENVA.01, to enhance immunogenicity.
