Ad35-GRIN is a recombinant, replication-defective adenovirus type 35 (Ad35) vectored vaccine candidate designed for the prevention of HIV-1 infection. Developed by the International AIDS Vaccine Initiative (IAVI) in collaboration with Crucell (now part of Janssen/Johnson & Johnson), the vaccine encodes a fusion protein consisting of four HIV-1 subtype A antigens: Gag, Reverse Transcriptase (RT), Integrase (Int), and Nef (collectively termed GRIN). The Ad35 vector was selected for its low naturally occurring seroprevalence in human populations, which helps to minimize the impact of pre-existing anti-vector immunity on vaccine efficacy. Ad35-GRIN is primarily designed to induce robust T-cell mediated immune responses, particularly CD8+ T-cell responses, against conserved regions of the HIV-1 virus. It has been evaluated in Phase I clinical trials, often as part of heterologous prime-boost regimens involving other vaccine modalities such as MVA-GRIN or adjuvanted protein vaccines.
