Ad35.CS.01

Ad35.CS.01 is a recombinant adenovirus serotype 35 (Ad35) vector-based vaccine expressing the full-length circumsporozoite (CS) protein of *Plasmodium falciparum*, minus the GPI anchor domain[2]. It was developed as a malaria vaccine candidate and evaluated in heterologous prime-boost regimens, typically as a priming dose followed by booster doses of RTS,S/AS01, another malaria vaccine targeting the same antigen[1][2][5]. The mechanism of action involves induction of both humoral and cellular immune responses against the CS protein; specifically, it aims to elicit CS-specific antibodies and polyfunctional CD4+ T-cell responses that may contribute to protection against malaria infection[6]. Clinical studies have shown that while Ad35.CS.01 can induce immune responses, its use as a prime in combination with RTS,S/AS01 did not significantly improve protective efficacy compared to three doses of RTS,S/AS01 alone[1][2][5].
