Ad4-EnvCN54

Ad4-EnvCN54 is a live, replication-competent recombinant adenovirus serotype 4 (Ad4) vector-based investigational vaccine designed to express a truncated HIV-1 clade C envelope glycoprotein (gp150, subtype CN54). The vaccine is administered orally in enteric-coated capsules and aims to induce immune responses against HIV. The Ad4 vector used as the backbone was originally developed from the wild-type virus employed by the US military for respiratory disease prevention. In this construct, the gene encoding HIV-1 Env (gp150) is inserted into the viral genome downstream of the E3 region. Upon administration, Ad4-EnvCN54 infects human cells and expresses membrane-bound trimeric Env protein to stimulate both mucosal and systemic immunity against HIV. The primary developer of this candidate is Imperial College London[3][6][7].
