Ad4-H5-Vtn is a recombinant, replication-competent adenovirus serotype 4 (Ad4) vectored vaccine expressing the hemagglutinin (HA) gene from the A/Vietnam/1194/2004 H5N1 influenza virus. It was developed as a mucosally-delivered vaccine candidate to induce protective immunity against H5N1 influenza. The Ad4 vector used is derived from the U.S. military Ad4 vaccine strain and has a partial deletion in the E3 region to accommodate the HA gene insert[2]. The vaccine induces both humoral and cellular immune responses, including increases in H5-specific CD4+ and CD8+ T cells, as well as IgG and IgA antibodies in serum and mucosal secretions[1][6]. Intranasal or upper respiratory tract administration leads to robust neutralizing antibody responses that are durable for at least 26 weeks post-immunization[1][7]. Preclinical studies demonstrated protection against lethal H5N1 challenge even with pre-existing immunity to wild-type Ad4[2]. Clinical trials have focused on evaluating safety, immunogenicity, optimal dosing, and delivery route for this platform[3].
