The Ad4-SARS-CoV-2 vaccine is an experimental viral vector vaccine candidate designed to elicit protective immune responses against COVID-19. It utilizes a replication-deficient adenovirus type 4 (Ad4) platform, typically engineered with deletions in the E1 and E3 genomic regions to prevent viral replication while providing space for the insertion of the SARS-CoV-2 spike (S) protein gene. Upon administration, the Ad4 vector delivers the spike gene into host cells, which then express the protein to stimulate both humoral (antibody) and cellular (T-cell) immunity. A significant area of research for this candidate is its potential for intranasal administration, which aims to induce mucosal immunity in the upper respiratory tract, potentially blocking viral entry and transmission more effectively than systemic injection. The Ad4 platform is of particular interest because it has been used safely for decades as an oral vaccine in military populations, and it may circumvent the high levels of pre-existing immunity found against other common vectors like Adenovirus type 5.
