Ad5-Δ24-RGD-BiTE is an engineered oncolytic adenovirus (OV) designed as a gene therapy platform to express a bispecific T-cell engager (BiTE). The viral backbone, based on Adenovirus type 5, features two key modifications for oncology: a 24-base pair deletion (Δ24) in the E1A region that restricts replication to cells with a disrupted Rb pathway (common in cancer cells), and an Arg-Gly-Asp (RGD) motif insertion in the fiber knob to enhance viral entry via integrins. This specific construct encodes a BiTE that simultaneously targets the T-cell marker CD3 and the tumor-associated antigen IL-13Rα2, which is highly expressed in glioblastoma. By combining direct viral-mediated oncolysis with the localized secretion of BiTEs, the therapy aims to overcome the immunosuppressive tumor microenvironment and recruit T cells to the tumor site, often used in synergy with CAR-T cell therapies.
